How to Read a Certificate of Analysis (COA)

A COA is only as credible as the lab that issued it. Supplier-issued COAs should be treated as a starting point, not a verification endpoint. Here is what to look for in any COA:

HPLC Purity >98%
Most research applications require >98% purity by HPLC. Some highly complex peptides may be acceptable at >95%. Anything below 95% warrants further investigation.
Mass Spec Confirmation
The COA should include an observed molecular weight matching the theoretical mass (within 1 Da or within 0.1% for large peptides). No mass spec = no identity confirmation.
Batch / Lot Number
Each COA must reference a specific batch number. A generic COA not tied to your specific lot is not a valid quality document for that sample.
Red Flags
No HPLC chromatogram, no mass spectrum, no batch number, COA issued by an entity with the same address as the vendor, or purity reported as a single number with no methodology.
Requesting Independent Testing
Contact a third-party lab directly. Provide compound name, expected MW, vial concentration, and desired tests. Retain a reference aliquot from the same vial you submit.
Test Date Currency
