PEG-MGF Protocol Guide
PEG-MGF (PEGylated Mechano Growth Factor) is a PEGylated form of the IGF-1Ec splice variant that is naturally produced in mechanically damaged muscle tissue. PEGylation extends the biological half-life from minutes to several hours, enabling systemic subcutaneous administration. MGF activates satellite cells (muscle stem cells), promoting their proliferation and fusion with existing muscle fibers for repair and hypertrophy. Unlike IGF-1 LR3 which drives differentiation, MGF specifically promotes the proliferative phase of muscle regeneration. This protocol covers 200mcg per muscle group post-training, cycling strategies, timing relative to workouts, and recovery stacking for optimal muscle repair research.
Protocol Overview
- Compound
- PEG-MGF (PEGylated Mechano Growth Factor / IGF-1Ec)
- Category
- Muscle Repair / Recovery
- Mechanism
- IGF-1 splice variant that activates satellite cell proliferation; PEGylation extends half-life for systemic delivery
- Half-Life
- ~15 minutes (MGF) / ~several hours (PEG-MGF)
- Form
- Lyophilized powder (2mg vials)
- Route
- Subcutaneous (near trained muscles)
- Frequency
- 2-3x weekly (training days)
- Cycle Length
- 4-6 weeks on, 4 weeks off
Dosing Protocol
| Protocol | Dose | Frequency | Route | Duration |
|---|---|---|---|---|
| Standard | 200 mcg / muscle group | 2-3x weekly (post-WO) | SubQ | 4-6 weeks |
| Bilateral | 200 mcg per side | 2-3x weekly (post-WO) | SubQ bilat. | 4-6 weeks |
| Recovery Focus | 200 mcg | 1x daily post-injury | SubQ near site | 2-4 weeks |
Key principle: Administer immediately post-workout for maximal satellite cell activation. Inject subcutaneously near (not into) the trained muscle groups. Do NOT co-administer with IGF-1 LR3 at the same time -- separate by at least 6-8 hours or use on different days.
Reconstitution Guide
Standard Reconstitution
- Vial: 2mg PEG-MGF lyophilized powder
- Diluent: 1mL bacteriostatic water
- Concentration: 2mg/mL (2,000mcg/mL)
- 200mcg dose: 10 units on insulin syringe
- Vial yield: 10 doses at 200mcg
Storage & Handling
- Pre-reconstitution: Freeze at -20°C for long-term storage
- Post-reconstitution: Refrigerate at 2-8°C
- Stability: Use within 21 days once reconstituted
- Avoid shaking: PEGylated peptides are sensitive to agitation
Expected Timeline
Side Effects & Monitoring
Common Side Effects
- Injection site soreness or swelling
- Temporary blood glucose fluctuations
- Mild lethargy post-injection (uncommon)
PEG-MGF is generally well-tolerated. Side effects are typically mild and transient.
Precautions
- Do not co-administer with IGF-1 LR3 simultaneously
- Monitor blood glucose, especially if diabetic
- Limited human clinical data available
- Avoid in subjects with active cancer (IGF pathway stimulation)
